Optische Plattform
Jeder Laserstrahl wird unabhängig ausgerichtet und verfügt zur optimalen Abstimmung über eine eigene Fokussierlinse.
Der 488-nm-Laser gehört zur Standardausstattung. Der 405-nm-Laser, der 561-nm-Laser und der 640-nm-Laser sind optional.

Emissionsoptik
Lenkoptik
Das Emissionslicht wird durch ein 20-faches, 0,6 NA-Mikroskopobjektiv gesammelt. Das Licht wird auf räumlich getrennte Spiegellochblenden fokussiert. Die gleichzeitige Videobeobachtung des Strahls, der Lochblenden und der Laserschnittpunkte ermöglicht eine schnelle und intuitive Ausrichtung.
Vorwärtsstreudetektor und Filter
Gesammelt mit 75- und 50-mm-Linsen und einer Photomultiplier-Röhre (PMT) mit einem 488/10-Bandpassfilter und einem OD3-Neutraldichtefilter.
Seitenstreudetektor und Filter
Durch die 90°-Sammellinse gesammelt und mit einem PMT mit einem 488/10-Bandpassfilter gemessen.
Fluoreszenzdetektoren und -filter
Durch die 90°-Sammellinse gesammelt und mit PMTs gemessen.
Fluidik
Allgemeiner Betrieb
- Das Fluidiksystem nutzt externen Luftdruck und Vakuum.
- Eine optionale Luftdruckversorgung und/oder Vakuumpumpe ist erhältlich.
- Der Manteldruck ist auf 27 psi (1,8 bar) festgelegt. Der Verbrauch an Mantelflüssigkeit beträgt etwa 0,5 l pro Stunde.
Fluidikbehälter
Im Lieferumfang sind autoklavierbare 7-Liter-Hüllen und Abfallbehälter mit Druck- und Vakuumanzeige enthalten.
Fluidik-Steuerung
- Zur präzisen Regulierung des Probenflusses ist eine Probenfluss-Feineinstellung vorgesehen.
- Zum schnellen Starten des Strahls und Entfernen von Blasen stehen die Funktionen „Entlüften“, „Pulsen“, „Spülen“ und „Ausführen“ zur Verfügung.
Düsen

Austauschbarer Fluidikpfad
- Der Fluidikpfad, einschließlich Düsenanordnung und Probenleitung, kann ausgetauscht werden. Es gibt keine Inline-Ventile. Es werden nur Quetschventile verwendet.
Blasendetektor
Ein Blasendetektor in der Probenleitung erkennt Luftblasen im Probenröhrchen und stoppt den Probenfluss, wenn das Probenröhrchen leer ist. Dadurch wird verhindert, dass Luftblasen die Düsenanordnung erreichen.
Beispieleingabe
- Das empfohlene Mindestprobenvolumen beträgt 0,2 ml. Das Totvolumen beträgt ca. 25 μl.

Die Verwendung der Integrator-Option ermöglicht die Unterscheidung von G2/M-Singuletts von G0/G1-Doubletts für PI-gefärbte Kalbsthymuskerne (CTN).

Vorwärts- und Seitwärtsstreuempfindlichkeit
Durch die Empfindlichkeit können fixierte Blutplättchen vom Rauschen getrennt und 1,0-μm-Kügelchen erkannt werden.

Durch die Streuauflösung ist eine Unterscheidung von Lymphozyten, Monozyten und Granulozyten möglich.

Reinheit und Ertrag
Bei 27 psi und 39 kHz mit einem durchschnittlichen Schwellenwert von 10.000 Ereignissen pro Sekunde erreichte eine Zweiwegesortierung eine Reinheit von 98 % und eine Ausbeute von >80 % der erwarteten Ausbeute von Poisson für beide Populationen. Höhere Schwellenwerte bis zu 20.000 Ereignissen pro Sekunde können ohne Beeinträchtigung der Reinheit erreicht werden; die Ausbeute wird jedoch aufgrund der Poisson-Statistik abnehmen.
Lebensfähigkeit
Sorten von Jurkat-Zellen führten zu >90% Lebensfähigkeit und ähnlichen Proliferationsraten * MESF
