High Precision Pump P-500
Introduction
The High Precision Pump P-500 is a laboratory pump for use in liquid chromatography and other applications where constant flow at pressures up to 4 MPa (40 bar , 580 psi) is required. Pump P-500 is very accurate and reproducible, especially at low flow rates over the whole pressure range. The excellent chemical resistance of the pump makes it possible to work with corrosive liquids, e.g. organic solvents, as well as aqueous solutions.
Pump P-500 is designed to facilitate routine chromatography work. The flow rate is easily and accurately set with a three digit thumb-wheel switch. The wide flow range makes the pump suitable both for analytical and preparative chromatography. A unique wash program provides fast and secure solvent change-over and a pressure Limit control protects the column from damaging pressures.
Together with the Gradient Programmer GP-250 Plus or the Liquid chromatography Controller LCC-501 Plus, the Pump P-500 can be used to form accurate and reproducible gradients.
Pump P-500 is fully compatible with other equipment such as the single and dual path UV-monitoring Systems UV-M 11, UV-1, and their associated recorders and with the Fraction Collectors FRAG-100, FRAC-200 and SuperFrac.
Pump P-500 is designed to work with a wide range of columns and gels supplied by Amersham Biosciences/Pharmacia Biotech.

HiLoad Pump P-50
Introduction
The liquid chromatography HiLoad® Pump P-50 is a robust biocompatible pump for use in liquid chromatography and other applications where constant flow rates up to 49.9 ml/min at pressures up to 0.5 MPa (5 bar) are required. Pump P-50 is accurate and reproducible over a wide range of flow rates, and is suitable for long-terrn use in dernanding environments.
Pump P-50 is designed for ease of use and minimum maintenance. The flow rate is quickly and accurately set with a 3-digit thumbwheel switch. A pressure sensor stops the pump if the back pressure exceeds 0.5+0.65 MPa, preventing damage to chromatography columns.
A remote control connector allows Pump P-50 to be used with Gradient Programrner GP-250 Plus, Liquid Chromatography Controller LCC-500 Plus, LCC-50i Plus or GradiFrac. Pump P-50 is also fully compatible with other liquid chromatography equipment such as UV-monitors UV-M II, UV-1 and Uvicord® Sil and fraction collectors FRAC-100, FRAC-200 and SuperFrac®.

Liquid Chromatography Controller LCC-500 Plus
Introduction
The Liquid chromatography Controller LCC-500 PLUS is a powerful, multi-functional control unit for chromatography systems. Its programmable memory allows you to control chromatographic procedures, modify separation parameters, perform multi-dimensional chromatography, evaluate and print-out data from the chromatogram. You can also control auxiliary equipment such as fraction collectors and Chart recorders at the saure time.
Fifty independent methods can be programmed and stored in the battery backed-up memory of the LCC-500 PLUS. Each method may be accessed individually, linked to other methods or called upon as
a submethod. In addition, methods or parts of methods may be looped up to 99 times. Programmed settings may be overridden manually at any stage.
Together with prepacked columns, pumps, valves, monitors, fraction collectors and automatic injector, the LCC-500 PLUS forms fully automated systems for fast, high-resolution liquid chromatography, such as FPLC® System and BioPilo System.

Method number
50 methods can be programmed individually. The methods are divided into 5 different method banks with 10 methods in each bank.
Memory capacity
750 instructions can be programmed if maximum peak memory is set to 20 (default). Otherwise every peak more than 20 will decrease the number of instructions available by 2 or 3. Minimum memory capacity is 569 instructions if 99 peaks are specified. Maximum memory capacity is 791 instructions if 2 peaks are specified.
Memory protection
The memory is backed-up with a lithium battery that will last for
a minimum of 5 years. A LED indicates when the battery should be replaced.
Display
16 digit alphanumeric display with 6 LEDs for status Information.
Keyboard
29 touch-type keys with audible tone acknowledgement. The tone will become louder when incorrect key entries are made.
Alarm
An audible alarm can be programmed to sound at any time (volume) with a duration of 0-99 min in increments of 0.1 min. There will also be a 30 s alarm when a check Code is indicated in the display.

Gradient formation
The gradient is formed by the control of either two pumps or one pump and a switch valve. Eluents are then mixed in a dynamically stirred mixing chamber. The supplied mixer has a volume of 0.6 ml, with magnetic stirring.
Concentration
0-100 %B in 0.1 % increments.
Gradient profiles
Any gradient profile can be programmed in linear Segments of different slopes. Linear gradients are programmed by entering the time (volume) and concentration of eluent B at the breakpoints. The concentration entered will be reached at the programmed time (volume). The concentration will be changed linearly between the breakpoints.
Data collection
Signals from 2 monitors within the input range 1 500 to +1 500 mV can be collected.
Peak detection
Two monitor inputs can be used for peak detection, but only one at a time. The value of the one not used for peak detection is however stored at peak maxima.
A peak is registered when the monitor input is above the set peak threshold level (% full scale deflection, FSD). Start of peak is the set level +2 or 2 % FSID above a minimum; end of peak is the set level 2, i.e. a hysteresis of 4.
Both peak monitor values and peak threshold level are referred to the current baseline of the respective monitor. The LCC-500 PLUS stores a new baseline value when the instruction MONITOR is executed.
lf , while running a method, the monitor value is below the programmed peak threshold level and slowly changing within the programmed maximal baseline drift, the LCC-500 PLUS updates and stores the monitor value as the new baseline.
When the monitor input is above the peak threshold level, the LCC-500 PLUS does not account for baseline drift, but refers all monitor inputs
to the baseline value stored at the moment the peak threshold level was passed. Baseline drift is then not accounted for until the monitor input is below the peak threshold level again.
Baseline drift is only compensated for if the threshold level is set to a non-zero value. A filter constant >2 must be programmed when the baseline function is used (for more Information about filter constants See section 4.2 "Calibration block").
Peak memory
Data from up to 99 peaks can be stored.
Peak evaluation
The following data from up to 99 peaks can be printed out:

Printer/plotter
The LCC-500 PLUS can control the built-in printer/plotter for listing of methods, calibration values and peak data. The printer can be made to plot and/or print data from the chromatogram by manual activation or through instructions in a programmed method.
If mains power failure occurs, a print-out containing method number, bank number, method time (volume) and retention time (volume) will be carhed out. The run will continue when the power returns.

Thermal paper length 25 m, width 60 mm, diameter max. 47 mm. Unprinted and printed paper should be stored in darkness in a cool, dry room.
A correct printing is only guaranteed when thermal paper type JUDO TP50 KS-F is used (Code no. 19-6828-01).
Programmahle outport
Seven programmable ON/OFF outports, e.g. for control of accessory equipment, TTL compatible. Three are designed for control of a fraction collector or automatic injector. The polarity for each outport can be programmed in the calibration block.
24 V Switched power outports
Three 24 V (max. 100 MA) switched power outports are available, of which two are designed to power solenoid valves etc and one for making one-pump gradients. The third is also useable as an outport for solenoid valves when using two pumps for gradierst formation. The power outports are powered down 30 ms alter any of the outports are activated to avoid overheating the valves.

Power failure/ disturbances
In the event of power failure the System will shut down. When the
power returns the run will continue exactly where interrupted and you will get a print-out (See Printer/plotter).

Valve control
The LCC-500 PLUS can control up to 6 motor-driven valves, MV-7, PMV-7, IMV-7, MV-8, PMV-8 and IMV-8. The allowed positions can be specifically addressed. If the addressed position is not found, an alarm together with a check code is given and the LCC-500 PLUS is paused.
MV-7, PMV-7, IMV-7
The MV-7, PMV-7 and IMV-7 can be addressed to their three positions LORD, INJECT and WASH. Addressing the valves to any other position will give rise to an audible alarm and a check code will appear in the display.
MV-8, PMV-8, IMV-8
The MV-8, PMV-8 and IMV-8 can be addressed specifically to any of their 8 positions. They can also be stepped forward (clockwise) to the next position.

Preprogrammed methods
The LCC-501 Plus has 9 different preprogrammed methods. The methods are within the Software and are copied into method bank 5 in method 0-8 by using the instruction CLEAR MEMORY 501 in the calibration block. If there already exists a method in one of these, this method will not be copied and check code 23 will be shown. There is one method for each technique (gel filtration, anion exchange, cation exchange, affinity, hydrophobic interaction and reversed phase chromatography) and three different equilibration methods.

Printer/plotter
The LCC-501 Plus can control the built-in printer/plotter for listing of methods, Calibration values and peak data. The printer can be made to plot and/or print data from the chromatogram by manual activation or through instructions in a programmed method.

Power failure/disturbances
In the event of power failure the System will shut down. When the power returns the tun will continue exactly where interrupted and you will get a print-out (See Printer/plotter).

Gradient Programmer GP-250 Plus
Introduction
The Gradient Programmer GP-250 Plus is a powerful, multi-functional control unit for chromatography systems. Its programmable memory allows you to control chromatographic procedures, rnodify separation parameters and control auxiliary equipment such as fraction collectors and chart recorders at the same time.
Fifty independent methods can be programmed and stored in the battery backed-up memory of the GP-250 Plus. Each method may be accessed individual !y , linked to other methods or called upon as a subrnethod. In addition, methods or parts of methods may be looped up to 99 times. Programmed settings may be overridden manually at any stage.
Together with prepacked columns, pumps, valves, monitors, fraction collectors and recorders, the GP-250 Plus forms a systems for fast, highresolution liquid chromatography, such as FPLC® System.

Fraction Collector FRAC-100 and FRAC-200
Introduction
The Pharmacia LKB fraction collectors, FRAC- 100 and FRAC-200 , are versatile Instruments for automated fraction collection and evaluation of chromatograms. In their standard forms they can collect up to 1.75 fractions in 12 mm diameter tubes, up to 95 fractions in
10-18 mm diameter tubes and up to 45 fractions in 28 mm diameter vials, with the help of the racks provided or with simple accessories. When connected to a suitable monitor, they can measure peak heights, areas and retention for up to 11 peaks (FRAC-100) or 19 peaks (FRAC-200). The fraction collectors will also indicate into which tubes the peaks haue been collected.
FRAC-200 and FRAC-100 (with its drop counting accessory) can collect fractions in a variety of different ways, based either an time, volume or drop counting, controlled via simple programs specified by the Operator. For example, you can choose to collect uniform fractions of a fixed time period. The fraction collectors will collect fractions until all the tubes are used and then switch off. In other collection sequences, you can specify a "wait" period in which the initial liquid volume is collected in a separate container, collect peaks in a smaller fraction size than the main pari of the eluent, collect only peaks and allow the remainder to run into a separate container, compensate for the liquid volume bet-ween the monitor and the fraction collector and program a time, volume or drop count for the collector to shut itself, and a connected pump, off automatically. At any time during collection you can advance to a new tube or stop the collection temporarily before continuing with the rest of the program.
These fraction collectors are designed to work with other equipment from Pharmacia LKB, including Peristaltic Pump P-1, Pump P-50, chromatography Controllers, Chart recorders and UV-monitors to form integrated chromatographic Systems adapted to your present and future requirements.

Introduction
Fraction Collector SuperFrac TM , is a sophisticated, high-capacity
preparative instrument enabling a wide variety of users to collect fractions or peaks according to their exact requirements.
To simplify operation an integrated microprocessor helps the user through the basic programming sequence. Optimal functions and parameters can be programmed independently.
SuperFrac fraction collector offers true peak-siope collection, time window collection, and many other advanced features. Peak-slope collection and time window collection are especially useful for repetitive preparative runs in lab scale, and pilot scale, where the ability of SuperFrac to accept many very large tubes is indispensable.

Introduction
Pharmacia Biotech model REC 101 (Code No. 18-1001-42), single-channel recorder and model REC 102 (Code No. 18-1001-43), dual-channel recorder are compact recorders designed to meet the challenging demands of todays technology.
These Instruments are part of the new generation of high quality, flat-bed recorders from Pharmacia Biotech which use a null balancing potentiometric system. They can accommodate a range of input voltages, offer a selection of chart speeds, and allow many functions to be remotely controlled.
The recorders have sophisticated features such as electric pen lift , grid and home functions, forward and reversed paper feed. Paper is transported at a constant velocity selected from the 14 different chart speeds in the range from 0.1 mm/min to 20 mm/s, and can be synchronized with the pump to enable chromatogram comparision.
The positions of the two independent pens (REC 102) represent the leveis of the two corresponding inputs from which the time functions are drawn.
Owing to the nature of the dual-channel recorder, the pen of channel 1 is always 2.5 mm behind the pen of channel 2. Full sensitivity ranges from 1 mV to 20 V DC.
Recorder (REC 102) is equipped with one chart drive control panel and two independent, isolated servo systems with a control panel for each channel.
Recorders REC 101 and REC 102 have an attractive design with much attention having been paid to ergonomics: control functions are weil positioned, keys and switches are very accessible, and paper is easily changed. Pens, paper and keyboard are all protected from accidental spillage by the moulded cover which fits over the recorder. The pens, which are nonsmear and disposable, give clear, accurate traces for instance evaluation. Chart paper is of a high quality, and 200 mm wide.

Conductivity Monitor
Introduction
The Pharmacia Conductivity Monitor, is a high quality, high precision on-line monitor for conductivity measurements and gradient determination in liquid chromatography applications.
The monitor consists of two units, the flow ccli and the control unit. The flow ccli contains the titanium electrodes used for conductivity measurement as weil as a temperature sensor. The small size of the flow ceil allows it to be positioned anywhere in the liquid path, provided that the pressure specification for the cell is not exceeded.
The control unit carries the six programming keys for the monitor. An easy to read 32 digit aiphanumeric display shows the conductivity measured in Siemens/cm (mS/cm or m S/cm) and the temperature. The display also shows the relative gradient Position as a percentage of the maximum set value, as both a percentage figure and graphically.
The monitor has a very large dynamic range from 1 m S/cm to 999.9 mS/cm and is therefore suitable for a wide range of applications, ranging from reversed phase to hydrophobic interaction chromatography. A unique design of the electronics is the microprocessor control of frequency variations within the working range. This gives excellent linearity and elirninates the need for range settings.

Monitor UV-M II
Introduction
The Pharmacia Monitor UV-M II , Hg Optics with 254 and 280 nm filters, Code No.18-0604-02 or Zn Optics with 214 nm filter, Code No. 18-0605-02) is a fixed wavelength monitor for liquid chromatography. The UV-M II consists of a control module and a small optical unit which makes it easy to place it optimally in the system. State of the art electronics and high precision optics offer fiexibility and accuracy in UV-M II detection. The UV-M II is ideal for all types of liquid chromatography, primarily FPLC® and HPLC.

The double cone shape of the analytical cell minimizes the collisions between light beams and cell wall and maximizes the illuminated volume, i.e. it gives an increased sensitivity. Easy selection of different filters means there is good choice for diverse application needs. This unit may be connected directly to any chromatography column, thus minimizing the dead volume between the column and the monitor.
The Control Unit has touch panel design and solid state electronics. Functions such as autozero and event mark can be executed from a remote source.

Introduction
Pharmacia Biotech Uvicord SII Code No. 18-1004-50 is a Compact single beam UV-monitor with flow-through cell for monitoring the eluant from various column Separations. Available wavelengths, selected by a combination of lamp and interference filier are 206, 226, 254, 280, 313, 365 and 405 nm.
Pharmacia Biotech Uvicord SII is housed up in one combined optical and electronic unit. This detector simultaneously produces two absorbance Outputs - one fixed at 2 AUFS and one selectable from 0.005 - 2 AUFS in 9 steps. Thus, with the selection of the appropriate absorbance Tange, Uvicord SII enables monitoring of both high and low concentration substances simultaneously with a dual-channel recorden
Substances such as non-aromatic peptides, amino acids, lipids, nucleotides and certain carbohydrates haue strong absorption at 206 nm whereas at higher wavelengths they Show little or no absorption. Furthermore, most proteins also exhibit greater absorption at 206 nm than they do at 280 nm.
