The API 2000TM LC/MS/MS System:
Triple Quadrupole Performance in a Compact Benchtop Footprint Featuring
Applied Biosystems /M DS SCI EX Ion Sources for Maximum Flexibility and Ease-of-Use
Overview
The API 2000TM LC/MS/MS System brings all of the power and performance of triple quadrupole mass spectrometry right to your benchtop . Other instruments claim to have a benchtop design - but use the entire bench. The API 2000 System leaves plenty of room for everything else in your lab. Its footprint measures a compact 45 cm X 100 cm.
In addition to the small footprint, the API 2000 LC/MS/MS System features robust ion sources from Applied Biosystems /MDS SCIEX. These sources offer full articulation with large viewing ports for easy optimization. To prevent contamination during switching, the sources
are positively vented and include the patented Curtain Gas TM interface to maximize ion transfer to the mass analyzer and provide superior ruggedness and reliability. Since safety is always a concern, the ion sources are fully insulated so that operators are protected from hot surfaces. Because time is so valuable, the ion sources for the API 2000 System are truly plug and play, thus allowing for a quick changeover between the sources to keep operations running smoothly without sacrificing productivity.
In general, triple quadrupole mass spectrometers are very popular analytical instruments. They offer many key benefits to researchers who are interested in high throughput analyte quantitation using a mass detector of high specificity coupled to a liquid chromatograph. Triple quadrupole MS/MS is one of the most sensitive and specific technologies available for small molecule quantitation . This technique is amenable to a broad range of compounds and inherently eliminates the need for extensive sample preparation. The fragmentation capabilities of tandem mass spectrometry also provide critical information for structural elucidation of unknown compounds.
Some typical applications for triple quadrupoles include:
- Drug Metabolism and Pharmacokinetics (DMPK)
- Environmental analysis
- Forensic toxicology
- Medical research
- Drug delivery
- Protein and peptide characterization
- Impurity and metabolite profiling
Key Features of the API 2000 LC/MS/MS System
- Small footprint
- LINACTM collision cell for high throughput Multiple Reaction Monitoring (MRM)
- Automated methods development
- Built-in syringe pump
- Economical
- Backed by a worldwide service and support network
Proven Performance
You wont have to sacrifice performance for size. The API 2000TM Systems proven engineering includes the proven Curtain Gas TM interface and the LINAC TM collision cell.
The latter allows for improved scan speeds and increased sensitivity while preventing contamination and eliminating cross-talk.
The Curtain Gas interface employs a nitrogen stream flowing countercurrent to the ion path. This gas flow drives solvents and buffers from the inlet, maximizing ion transfer to the analyzer, thereby providing unmatched ruggedness and reliability. Efficient Ionization is achieved without heat. This eliminates the possibility of thermal degradation of labile compounds.
The Gold Standard in Ionization Performance
Applied Biosystems /MDS SCIEX offers two ion sources for the API 2000 System to accommodate all of your analytical demands:
Heated Nebulizer or Atmospheric Pressure Chemical Ionization (APCI) Source
- Full gradient capabilities
- Computer controlled probe temperature and gas flow
- Optional Supercritical-Fluid Chromatography (SFC) compatibility
- Flow rates up to 2 mL per minute
- Allows ionization of neutral molecules
- Species are less susceptible to ionization suppression
- Wide dynamic range
- LC dead volume is less of a consideration
- Higher buffer concentrations are acceptable
TurboIonSpray TM Source
- Permits flow rates from 2 µL to 1 mL per minute
- Exhibits excellent concentration dependent response
- Ability to analyze high molecular weight compounds
- Very sensitive
- Wide range of accepted analytes
- Experiences none of the problems associated with thermal lability or volatility
Ease-of-Use
Never before has an LC/MS/MS system been so user friendly. The APCI and TurboIonSpray sources can be completely interchanged within minutes without tools. The following three steps illustrate the ease with which the sources can be switched
.
All temperature, gas and electrical connections are fully integrated into the source housing so there are no extra lines to attach. This seamless conversion is completed when magnetic connections automatically detect and indicate the hardware change to the software. There is no need to tell the software what source you are using. The built-in syringe pump and diverter valve simplify hardware configuration and also contribute to conserving bench space.
Superior engineering allows for reliable flow rates as low as 2 µL per minute and up to 2 mL per minute. In addition, ion sources are safety interlocked - voltages are automatically set to zero when either source is removed.
All mass spectrometers require occasional cleaning and maintenance. Maintenance is simple with the API 2000TM System. The system covers are easily removed with common tools, and components such as the curtain plate, orifice, skimmer, and entrance quadrupole are readily accessible for cleaning.
The API 2000 System also features Analyst TM software for acquisition, instrument control and data processing. Analyst software simplifies data reduction in the familiar Windows NT® operating environment. Automatic tuning and calibration ensure optimum performance while rapid data acquisition increases your productivity, allowing you to spend valuable time exploring and interpreting data instead of on system set-up. Analyst software further streamlines your system by offering single point control over a wide selection of LC pumps, autosamplers and liquid handling systems.
Conclusions
Laboratories of all sizes can benefit from the advantages of tandem mass spectrometry with the API 2000 LC/MS/MS System. This rugged system gives you the high throughput and specificity that you need in an economical, benchtop platform. Productivity is improved even further with the API 2000 System because of its rugged and easy to use ion sources. The Curtain GasTM interface and LINACTM collision cell are instrumental in providing superior
performance . Experience the benefits of a triple quadrupole instrument with a smaller footprint and less maintenance than you would expect.

Series 200 Vacuum Degasser
About the Series 200 Vacuum Degasser
The PerkinElmer Series 200 Vacuum Degasser is a high- efficiency, online module that removes dissolved gasses from HPLC solvents. Its unique design assures reliable continuous operation and the highest level of continuous performance available without the need for helium degassing. The five-channel design is compatible with isocratic, binary, and quaternary pumping systems, and includes an additional channel to provide a separate degassed solvent to the autosampler needle wash port. A three-channel design is also available and is compatible with binary pumping systems that include an autosampler . The figures shown in this manual are for the quaternary version of the degasser. The multi-pass design, (five channels) allows increased flexibility in that you can connect one quaternary pump and an autosampler , two binary pumps and an autosampler or four isocratic pumps and an autosampler . Also, this design permits the use of higher flow rates by passing the solvents through the chambers several times. The extremely low internal volume of each Teflon AF ® channel (400 microliters ) offers very quick equilibration and very short startup times compared with PTFE degassing channels which have the same degassing efficiency.
Inside the unit, the solvent flows through a short length of Teflon Ar tubing which has a volume of 400 microliters in a vacuum chamber. Within this chamber a partial vacuum is maintained, i.e. a pressure below 54 mm of Mercury (Hg) absolute by a constantly running, low RPM vacuum pump. Dissolved gasses are pulled through the tubing wall by the vacuum as the solvent flows within the coil, in accordance with Henry's Law. Gasses removed are expelled, and the chamber is maintained at the maximum vacuum level attainable by the vacuum pump.
A special port in the vacuum pump continually flushes the pump head with a small "bleed" of air to remove any solvent vapors that may enter the pump from the vacuum chamber. This air bleed eliminates the need for any solenoid valves within the system. This design results in zero vacuum " hysteresis ". Previous designs allowed the vacuum chamber pressure to fluctuate, with the pump cycling an and then off in response to the vacuum level. A patent is pending an this design concept.
Series 200 LC Pump
