INTRODUCTION

Multiskan PLUS is an eight-channel vertical light path filter photometer designed to measure the absorbances of liquids in flat-bottomed, 8x12 well microplates, and flat-bottomed Microstrips® of 1x8, 3x8 or 1x12 wells.

The wavelength (400-750 nm) is selected using high quality interference filters.

Four measurement modes and eight calculation modes are included. Any of the calculation modes can be applied to the results of any of the measurement modes.

Multiskan PLUS is designed to be used in various colorimetric and turbidimetric measurements typical of enzyme immunoassay (EIA) and micro­biological work.

PRINCIPLE OF OPERATION

The optics of Multiskan PLUS are based an Labsystems" concept of vertical photometry in which the light beam passes through the whole sample.

In vertical photometry, the absorption of light is proportional to the amount of light absorbing substance in the well.

Absorption is expressed by the following equation:

A = a/S m,

where A = absorbance, a = molar absorptivity of the substance, m = mass of absorbing substance and S = cross-sectional area perpendicular to the light path.

The advantages of the vertical light path measurement are:

(1) inaccurate pipetting of nonabsorbing liquids does not affect the measured absorbance values

(2) evaporation of nonabsorbing liquids during the reaction does not affect the measured absorbance values and

(3) a certain degree of inhomogeneity in the solution, for example as a result of layering in turbidity measurements, does not affect the results.
