Dispatch time

Das Sysmex R-2000 arbeitet nach dem Prinzip der Durchflußzytometrie. Nach Ansaugen der Probe werden 10 µl Blut im Probendosierventil abgeschert, mit Pufferlösung (RETSEARCH buffer) 1:200 verdünnt und mit dem Supravitalfarbstoff Auramin-O (RETSEARCH dye) angefärbt. Ein Aliquot dieser Suspension wird anschließend in einer Durchflußküvette hydrodynamisch fokussiert (RET-SHEATH) und durch den Strahl eines Argon-Lasers belich­tet.

Die durch den Farbstoff Auramin-O angefärbten Nukleinsäuren der Zellen erzeugen eine Fluoreszenz, deren Stärke vom Nukleinsauregehalt abhängt. Diese Fluoreszenzsignale werden aufgezeichnet. Gleichzeitig wird von jeder einzelnen Zelle das Vorwärtsstreulicht als Maß für die Zellgröße registriert.

Aus beiden Signalen wird das Scattergramm erstellt, in dem jede einzelne Zelle mit einem ihrer Größe (y-Achse) und ihrem RNA- bzw. DNA-Gehalt (x-Achse) entsprechenden Punkt eingetragen ist. Zwei vom System individuell für jede Probe gesetzte Diskriminatoren trennen die Retikulozytenpopulation von den Erythrozyten und den Thrombozyten. Eine weitere Schwelle trennt nach rechts die Normoblasten und Leukozyten von der Retikulozytenpopulation ab. Die Alters­klassifizierung der Retikulozyten wird an­hand ihrer Fluoreszenzintensität vorgenom­men.

Drei Klassen werden gebildet:

Zellen mit geringer (LFR),

mittlerer (MFR) und

ho­her (HFR) Fluoreszenzintensität.

Die Meßwerte werden als prozentualer Anteil der Gesamtretikulozytenzahl angegeben.
