Introduction to the 2D 2920 Master Imager

The 2D 2920 Master Imager is a multi-wavelength fluorimager capable of fluorescence measurements on 2D protein gels. It has a maximum scanning area of 240 X 280mm at resolutions of up to 50 microns. Fluorescence measurements may be made using any combination of up to six excitation and emission wavelengths, within the range 400 to 700 nm. The 2D 2920 Master Imager is supplied with a high specification PC, and is extremely easy to use via its Windows 95 TM compatible software and multi-format image export capabilities.

The protein gel is placed horizontally on the carrier plate, which is automatically loaded into the instrument when a scan is requested. A cooled CCD camera scans the required area capturing a number of FRAMES, which are combined to produce an IMAGE of the fall gel. Each frame is of fixed size (approximately 50 X 70 mm).

A high resolution mosaic image (an image made up of frames) can be produced by either edge or top illumination of the protein gel using a high power xenon light source with interference filters for fluorescence measurements. The edge illumination light guide is fixed in position at the rear edge of the sample. The top illumination light guide moves with the camera. The edge illumination method is especially applicable to gels where it distributes the available light between the glass plates. The software enables the illumination method and wavelengths to be set to capture multiple images of the CyDye TM labelled protein samples present on the same gel.

Gel images may then be subjected to detailed analysis within the 2D Master software package or exported in a variety of standard fite formats for more specific analysis using the Image Master TM Software.
