DER DNA THERMAL CYCLER

Der Perkin-Elmer/Cetus DNA Thermal Cycler wird zur Automation des Polymerase-Kettenreaktionsverfahrens (PCR) zur DNA-Verviel-fältigung eingesetzt. Der Mikroprozessor gibt Ihnen die Möglich-keit, entsprechend Ihrem Ausgangsmaterial und Ihren spezifischen Forschungszwecken die geeigneten Programme für die Eingabe von Zeit- und Temperaturparametern zu wählen. Diese Programme können in Files abgelegt werden, die für weitere Reaktionsverfahren abgeändert werden können. Zusätzlich können mehrere Files seriell miteinander verknüpft werden, wodurch die Durchführung kom-plexerer Vervielfältigungsprogramme ermöglicht wird.

Bei dem DNA Thermal Cycler handelt es sich um einen pro-grammierbaren Probenhalterblock zum Aufheizen/Kühlen der Proben. Der Block enthält 48 Bohrungen zur Aufnahme der Reaktionsgefäße (0,6 ml Inhalt). Mit Hilfe des Tastenfeldes und der Bildschirmanzeige können die Benutzerprogramme (Files) generiert, geändert und überarbeitet werden. Weiterhin können die verschiedenen Parameter während des laufenden Gerätebetriebes überprüft werden, da die aktuellen Temperaturen und Zeiten konti-nuierlich angezeigt werden. Für die Generierung, Verknüpfung und den Ablauf der Dateien stehen acht Funktionstasten zur Verfügung. Auf dem numerischen Standard-Tastenfeld werden Zeit- und Temperaturparameter, Kennummern des Benutzers und Datei-nummern eingegeben. Die Leuchtanzeige auf dem Bildschirm (40 Zeichen) erscheint während des Generierens der Dateien und liefert während des Betriebes Informationsdaten der aktuellen Methode. Die Kontrollampen für den Heiz- oder Kühlvorgang auf der linken Seite des Tastenfeldes zeigen an, ob der Thermal Cycler gerade heizt oder kühlt. Die Anzeige "Hot" leuchtet auf, sobald die Blocktemperatur 50 eC überschritten hat.
