Principle of operation

The radiation coming from the mercury-xenon lamp is converged and enters the excitation monochromator, in which it is dispersed by the large-aperture concave grating, and only the light beam of a specific wavelength band is allowed to pass through the exit slit. The light beam passing through the excitation monochromator ir­radiates the Sample through the condenser lens. Thus, fluores­cence is emitted from the Sample. The fluorescing radiation from the sample enters the emission monochromator, in which it is dis­persed by the concave grating, and the selected light beam is given to the measuring photomultiplier detector.

On the other hand, the excitation light beam transmitted through the sample reaches the monitor detector through the condenser lens, attenuator and diffusion plate. The voltage applied to the meas­uring photomultiplier is controlled using the output of this moni­tor detector. Therefore, even if the intensity of radiation from the light source fluctuates, correction is made by changing the gain of measuring photomultiplier.
