Introduction
The most popular liquid chromatographic detectors are those which operate on physical principles, such as absorption or emission of light or refractive Index changes. Of these, luminescence detectors are the most sensitive and selective, with fluorescence detectors particularly popular. The techniqu, can be applied to detect the native fluorescence of many organic molecules as well as the emission from fluorescently-labeled derivatives of non-fluorescing -fluorescin compounds. Indeed with improved autosampling devices, chemical derivatization with fluorescent reagents can be perfornied online immediately before the column. For the best results, the detector should be equipped with gratings before and after the flow cell for programming of excitation and emission wavelengths. This, among other things, is one of the features that makes the Hem-lettPackard HP 1046A fluorescence detector the ideal choice for highly sensitive work. The detec. tor can be added to an existing liquid chromatograph as a standalone unit or integrated in an automated HPLC System, with integrator control or PC-based method programming on the Hewlett-Packard HPLC ChemStation.
