Use or Function
The Iuminescence analyzing system is a fully automatic open analyzer for the in-vitro diagnostics.
Its advantages are:
Module construction with automatic rack transport from the pipetting station to the measuring unit.
Capacity to handle up to 280 samples per loading.
Automatic pipetting of all samples using up to 4 assays one after the other.
Automatic incubation of all samples (vibrating at room temperature).
Automatic washing of all samples in blocks of 10.
Open system, suitable for supplier's standard kits: LIA, ILMA, SPALT, using coated beads or coated tubes.
Highly sensitive photomultipliers for measuring the luminescence reaction by photon counting.
The LIA-MAT SYSTEM 300 system uses transport racks and can hold 10 test tubes measuring 12 x 75 mm.
In the luminometer system two starter reagents or analyzer reagents are automatically added which set off the Iight reaction. Then the samples are measured and the reagents are aspirated.
The AT compatible processor can process a variety of measuring and control procedures. Additionally, each module contains its own intelligence, that means a microprocessor which can independently process the presented task (multi-tasking).
A standard curve can be calculated from the measurement results or the standard sera of the immunoassays.
The unknown concentrations of the patients sera/control can be calculated from this curve.
Principle of Luminescence
The luminescence is a reaction during which light (photons) is emitted.
A molecule is stimulated by absorbing chemical energy. From this stimulated condition the molecule goes back to a lower level by emitting Iight of a defined wave length.
We distinguish between bioluminescence and chemiluminescence:
Bioluminescence are the light phenomena of living organisms Ilke the glow-worm, the American firefly or photo bacteria. By oxidation the production of Iight is catalyzed by specific enzyme systems.
The chemiluminescence has the same physical basis, but no living organisms are involved. The Iight-producing molecules are chemical compounds which can be activated by oxidation. They can release their stored energy within shortest intervals of time in a light-emitting reaction. The Iight reaction is not spontaneous but can be set off at a specified point of time by injecting a starter reagent.
Before this injection takes place, a catalyst or an acid has to be added.
The emitted photons hit the multiplier and are there integrated (counted) during the measurement time. The unit of the light mass is RLU (relative light units).
The most frequently used luminogeneuous compounds are:
- acridinium ester I salts
- aminophthal hydrazide
The luminescence immunoassays are non-isotopical immunochemical processes for the laboratory diagnostic whose antibodies and antigenes are Iabeled luminogeneuously.
The Iuminescence assays are in their sensitivity and specification comparable with radioactive assays, but oller the advantages in handling and disposal since they are not labeled radioactivity.
Similar to the radioactive assays, different types of assays are distinguished:

SPALT solid phase antigen Iuminescence technique
Depending whether it is a competitive or a sandwich assay, the results are falling or rising standard curves.
In any case, the Iight mass is proportional to the amount of luminogeneuous substance (tracer) in or at the test tube.
The released Iight mass therefore correlates with the concentration of the tested analytes.
